Inspiré par la capacité de réduction spéciale de l’acide ascorbique (AA), le 2-phosphate d’acide ascorbique (AA2P) a été largement utilisé comme substrat dans les tests actuels d’activité de la phosphatase alcaline (ALP) en raison de la transformation de l'AA2P déclenchée par ALP en AA. Cependant, de tels tests nécessitent généralement des procédures de synthèse et / ou de génération de signal compliquées et laborieuses liées aux AA. Nous rapportons ici une interaction fluorogénique in situ intéressante entre l'o-phénylènediamine (OPD) et l'AA, qui nous incite à proposer pour la première fois un nouveau et simple test d'activité d'activation de la ALP par fluorescence à la participation de AA2P / OPD Le test immuno-absorbant lié à une enzyme de fluorescence (ELISA) correspondant à base d'ALP a également été mis au point au moyen des plateformes classiques ELISA. Selon le processus de détection pratique et facile avec un mécanisme de réponse clair, notre test basé sur une réaction fluorogénique présente une bonne sensibilité, une sélectivité et une excellente performance de détection, ce qui garantit que le test ELISA de fluorescence peut potentiellement être appliqué au diagnostic clinique en utilisant un biomarqueur bien étudié du virus hépatocellulaire. carcinome, alpha-foetoprotéine (AFP) comme analyte modèle.
