L'acide ascorbique encapsulé dans des liposomes chargés négativement présente une perméation cutanée accrue, une rétention accrue et une synthèse accrue du collagène par les fibroblastes.

L'acide ascorbique (AA) est largement utilisé dans les formulations cosmétiques en raison de ses propriétés antioxydantes et de sa capacité à augmenter la synthèse de collagène. Ici, nous avons encapsulé AA dans des vésicules avec différentes compositions lipidiques. La charge liposomique négative a favorisé la rétention cutanée des AA, avec une accumulation de 37 ± 12 et de 74 ± 23 mug / cm (2) dans l'épiderme et le derme, respectivement, au bout de 6 heures. Le flux de médicament était influencé par la composition de la formulation, et la présence de cholestérol et la charge de surface des liposomes ont pu augmenter la quantité d'AA traversant la peau. La formulation était stable pendant au moins 30 jours et favorisait une augmentation du flux de 7 fois supérieure à celle de l'AA libre. De plus, les liposomes ont pu mieux interagir avec les membranes des kératinocytes et des fibroblastes. Des études d'efficacité in vitro ont montré que l'association de l'AA à ces liposomes entraînait une efficacité accrue de la synthèse du collagène de type I par les fibroblastes et la régénération des lésions induites par les UVA dans les kératinocytes. Nos résultats démontrent l'applicabilité des liposomes chargés en AA dans la promotion de la perméation cutanée à l'AA et l'augmentation de la rétention et du flux de cette molécule dans la peau. Cette formulation augmente également la stabilité et l'efficacité des AA, ouvrant de nouvelles perspectives pour son application en vue de réduire certains effets du vieillissement de la peau.
