Les protéines TET (Ten-Eleven Translocation) catalysent l'oxydation de 5mC (5-méthylcytosine) en 5hmC (5-hydroxyméthylcytosine) et jouent un rôle crucial dans le développement embryonnaire. L'acide ascorbique (Vc, Vitamine C) stimule l'expression des protéines TET, alors que le DMOG (diméthyloxallylglycine) inhibe l'expression de TET. Pour étudier le rôle de TET1, TET2 et TET3 dans le développement embryonnaire de PA (parthénogénétique), des traitements Vc et DMOG ont été administrés au cours du développement embryonnaire précoce. Les résultats ont montré que le traitement par Vc augmentait le taux de blastocystes (20,73 ± 0,46 par rapport à 26,57 ± 0,53%). En revanche, le DMOG a réduit le taux de blastocystes (20,73 ± 0,46 par rapport à 11,18 ± 0,13%) chez les embryons de PA. Les résultats de la coloration qRT-PCR (PCR en temps réel quantitative) et IF (immunofluorescence) ont révélé que les expressions de TET1, TET2 et TET3 étaient significativement plus faibles chez les embryons PA que chez les embryons fertilisés (Con) normaux. Nos résultats ont révélé que Vc stimule l'expression des protéines TET dans les embryons de PA. Cependant, le traitement au DMOG inhibe de manière significative l’expression des protéines TET. De plus, 5hmC ont augmenté après le traitement par Vc et supprimés par DMOG chez des embryons de PA. Pris ensemble, ces résultats indiquent que l'expression des protéines TET joue un rôle crucial joué par 5hmC dans le développement embryonnaire du PA.
