L'acide ascorbique (AsA) est un cofacteur enzymatique et antioxydant important dans de nombreux processus biochimiques. La plupart des activités biologiques de l’AsA sont étroitement liées à ses propriétés rédox. Des études récentes ont démontré que l'AsA est associé à des maladies liées à l'amyloïde et peut inhiber l'agrégation de polypeptides par l'amyloïde. Dans la présente étude, nous avons déterminé la cinétique de la dégradation de l'AsA et étudié les activités anti-amyloïdogènes de l'AsA et ses produits de dégradation en utilisant l'insuline comme polypeptide modèle. Les résultats ont montré que la demi-vie de l'AsA variait avec le pH du milieu et la température d'incubation. Les produits de dégradation de l'AsA ont inhibé la fibrillation d'insuline, avec une activité corrélée positivement au degré de dégradation de l'AsA. L'espèce de dégradation, comparée à l'As intacte, a également montré un effet perturbateur plus fort sur les fibrilles amyloïdes matures et une diminution significative de la cytotoxicité fibrillaire. L'acide déhydroascorbique et l'acide dicétogulonique, deux intermédiaires clés de la dégradation de l'AsA, avaient une activité anti-amyloïdogène similaire vis-à-vis de l'espèce de dégradation de l'AsA. Les résultats de ces travaux indiquent que la dégradation des antioxydants naturels doit être prise en compte lors de l'évaluation de leurs effets anti-amyloïdogènes. Ces informations sur l'action de l'AsA pourraient également fournir une nouvelle voie pour comprendre ses rôles physiologiques / pharmacologiques dans les maladies liées à l'amyloïde.
