Les antioxydants naturels et les vitamines ont le potentiel de protéger les systèmes biologiques des dommages peroxydants induits par les radicaux peroxyle, l’alpha-tocophérol (vitamine E, soluble dans les lipides) et l’acide ascorbique (vitamine C, solubles dans l’eau), molécules bien connues des antioxydants naturels. Dans la présente étude, nous avons décrit la synthèse et l'évaluation biologique d'hybride de ces deux antioxydants naturels via un lieur diéthylique d'ammonium, Toc-As dans la délivrance de gènes. Deux lipides cationiques témoins, N14-As et Toc-NOH, sont conçus de manière à ce que l'un se trouve avec une fraction acide ascorbique et aucune fraction de tocophérol; une autre est avec une fraction de tocophérol et aucune fraction d'acide ascorbique respectivement. Les trois lipides cationiques peuvent former des agrégats auto-assemblés. Les rendements antioxydants des trois lipides ont été comparés à ceux de l’acide ascorbique libre. Les lipides cationiques (Toc-As, N14-As et Toc-NOH) ont été formulés individuellement avec un co-lipide fusogène bien connu DOPE et des études de caractérisation telles que la liaison à l'ADN, le déplacement de l'héparine, la taille, la charge et le dichroïsme circulaire ont été réalisées. Les études de caractérisation biologique telles que le test de viabilité cellulaire et les études de transfection in vitro ont été réalisées avec les formulations ci-dessus dans les lignées cellulaires HepG2, Neuro-2a, CHO et HEK-293T. Les trois formulations ont montré leurs efficacités de transfection avec le plus élevé en Toc-As, modéré en N14-As et le moins en Toc-NOH. Fait intéressant, l'efficacité de la transfection observée avec le lipide conjugué à base d'antioxydant Toc-As s'avère être environ deux fois et demie plus élevée que la lipofectamine 2000 disponible dans le commerce à un rapport de charge de 4: 1 dans les lignées cellulaires Hep G2. Dans les autres lignées cellulaires étudiées, l'efficacité de Toc-As s'est avérée supérieure ou comparable à celle de la lipofectamine 2000. Les résultats de la caractérisation physico-chimique montrent que le lipide Toc-As montre un potentiel antioxydant maximal, une forte liaison avec le pDNA, potentiel zêta optimal. Il s'avère également être le moins toxique dans toutes les lignées cellulaires étudiées, en particulier dans les lignées cellulaires Neuro-2a, par rapport aux deux autres lipides. En résumé, le lipide antioxydant conçu peut être exploité en tant que système d'administration pour traiter des maladies associées aux ROS telles que la malignité, les accidents cérébrovasculaires, etc.
