Contexte: Les poules pondeuses de plus de 75 semaines présentent généralement une baisse importante de l'immunité et des performances de production. Il n'est pas clair si ces baisses peuvent être atténuées en ajoutant de l'acide ascorbique (AA) dans les aliments. Deux essais ont été menés pour étudier la synthèse et le métabolisme de l'AA dans des couches d'âges différents et les effets d'un supplément alimentaire en AA sur les performances et le statut immunitaire et antioxydant de poules âgées de 78 semaines. Méthodes: En Exp. 1, un nombre égal (24 poules) âgés de 35 semaines (jeunes) et de 75 semaines (vieux) ont été nourris au même régime sans supplémentation en AA pendant 4 semaines. En exp. Deux à 360 poules pondeuses en bonne santé âgées de 78 semaines ont été assignées au hasard à 4 traitements (régime de base complété de 0, 0,25, 0,5 ou 1 g AA / kg de diète) dans un essai d'alimentation de 8 semaines. Résultats: Les poules âgées avaient tendance à avoir une activité diminuée de la L-gulonolactone oxydase (GLO) synthase dans les reins et le foie par rapport à celle des jeunes poules (P = 0,07 et P = 0,05, respectivement). Comparées aux jeunes poules, les poules âgées présentaient une capacité antioxydante hépatique inférieure permettant l'expression génique de la thiorédoxine inférieure (TXN), de la thiorédoxine réductase (TXNR) et du cytochrome b5 réductase (CYB5R) (P <0,05), tandis qu'une augmentation de la vitamine C dépendant du sodium transporteur (SVCT) 1 niveaux d'expression dans l'iléon et le rein et augmentation des concentrations en AA splénique et hépatique (P <0,05). La supplémentation alimentaire en AA diminue de manière significative l'activité enzymatique des GLO mais augmente la concentration en AA splénique et les taux d'IgG anti-albumine sérique bovine (P <0,05) et tend à augmenter le nombre de lymphocytes T CD4 (+) (P = 0,06) dans le sérum. Une supplémentation de 0,25 g AA / kg de diète a significativement augmenté la capacité antioxydante totale hépatique (T-AOC, P <0,05) par rapport au groupe témoin. Conclusions: Les poules pondeuses pourraient synthétiser les AA dans les reins et le foie, bien que les activités de l'enzyme GLO soient 100 fois plus importantes dans les reins que dans le foie. Les vieilles poules pondeuses avaient une plus grande capacité d'absorption et de réabsorption et une plus grande rétention d'AA dans certains tissus que les jeunes poules. La supplémentation alimentaire en AA peut améliorer la santé des anciennes couches en renforçant l'immunité et la capacité antioxydante.
