L'acide L-ascorbique (vitamine C, AA) présente des effets anticancéreux avec un traitement à forte dose via la génération d'espèces réactives de l'oxygène (ROS) et des lésions sélectives des cellules cancéreuses. Les effets anticancéreux de l'acide L-ascorbique sont déterminés par le transporteur de vitamine C dépendant du sodium 2 (SVCT-2), un transporteur de l'acide L-ascorbique. Dans cette étude, nous démontrons que le traitement à l'acide L-ascorbique a montré une activité anticancéreuse efficace dans des lignées cellulaires avec des niveaux d'expression élevés de SVCT-2 pour une concentration de gradient d'acide L-ascorbique à partir de 10 µM -2 mM. Cependant, dans les lignées cellulaires à faible expression de SVCT-2, l'acide L-ascorbique à forte dose (> 1 mM) présentait des effets anticancéreux mais un traitement à faible dose (<10 μM) induisait une prolifération cellulaire. Ces résultats contradictoires qui dépendent de la concentration sont appelés une réponse à une dose hormétique. Une réponse de dose hormétique à l'acide L-ascorbique à faible dose a également été observée dans des lignées cellulaires à forte expression de SVCT-2 en présence d'un inhibiteur de la famille des SVCT. L'absorption insuffisante de l'acide L-ascorbique dans les lignées de cellules cancéreuses à faible expression de SVCT-2 ne peut générer suffisamment de ROS pour tuer les cellules cancéreuses, entraînant ainsi une réponse hormétique. L'analyse moléculaire a confirmé l'augmentation de l'expression des marqueurs de prolifération du cancer dans la réponse aux doses hormétiques. Ces résultats suggèrent que L-ascorbic présente un effet biphasique dans les cellules cancéreuses en fonction de l'expression de SVCT-2.
