Objectif: étudier l'effet protecteur de l'acide ascorbique (AA) sur les lésions des fibroblastes embryonnaires de souris exposés au nickel (NIH / 3T3). Méthodes: Un modèle de dommage induit par 50 mug / mL de poussière de raffinage de nickel a été établi afin de déterminer le taux de survie relatif des cellules, la superoxyde dismutase (SOD), la lactate déshydrogénase (LDH) et la glutathion peroxydase. (GSH-Px), teneur en peroxyde d’hydrogène (H (2) O (2)) et en malondialdéhyde (MDA) et expression de la protéine p53 (de type sauvage) Bcl-2. Étudier l'effet protecteur de différentes doses d'acide ascorbique (25, 50, 100 mmol / L) sur les lésions des cellules NIH / 3T3 induites par la poussière raffinée au nickel. Résultats: L'étude a montré que le groupe acide ascorbique peut augmenter considérablement le taux de survie des cellules NIH / 3T3; Le taux d'apoptose a été réduit; La vitalité de la SOD et de GSH-Px a augmenté de manière significative et la différence était statistiquement significative (p <0,05). Dans le même temps, les taux de MDA et de H (2) O (2) et l'activité de l'enzyme LDH extracellulaire étaient significativement réduits et la différence était statistiquement significative (p <0,05). Les résultats ont montré que la poussière de raffinage du nickel induisait des dommages cellulaires dus à la régulation à la hausse de la protéine p53 et à la régulation à la baisse de l'expression de la protéine Bcl-2; interventions d’acide ascorbique, le niveau d’expression de la protéine Bcl-2 dans les groupes d’acide ascorbique II et III était supérieur à celui du groupe de poussières de raffinage du nickel, et la différence était statistiquement significative (P <0,05); Le niveau d'expression de la protéine p53 dans chaque groupe de dose d'acide ascorbique était inférieur à celui du groupe de poussières raffinées au nickel et la différence était statistiquement significative (p <0,05). Conclusion: avec l'augmentation de la concentration d'acide ascorbique, les niveaux de dommages oxydatifs, les niveaux d'enzymes antioxydantes, la réduction de l'apoptose cellulaire, la réduction de l'expression de p53, l'augmentation de l'expression de Bcl-2. Il a montré que l'acide ascorbique avait un effet protecteur sur les dommages causés aux cellules NIH / 3T3 par la poussière de raffinage du nickel.
