Le carcinome thyroïdien anaplasique (ATC) nécessite des approches plus innovantes, les régimes thérapeutiques actuels étant inadéquats, la plupart des médicaments anticancéreux entraînant également une suppression générale des fonctions physiologiques. Cependant, un traitement avec des lésions tissulaires non ciblées limitées est souhaitable. Dans la présente étude, nous montrons le pouvoir prooxydant de l’acide ascorbique, qui améliore la cytotoxicité induite par la juglone. Nous décryptons que la combinaison juglone-ascorbate induit une apoptose induite par les espèces réactives de l'oxygène conduisant à la mort cellulaire dans la lignée cellulaire ARO issue de l'ATC. Cette combinaison affecte également l'activité enzymatique de la balance rédox déstabilisante déstabilisante déstabilisante de la catalase, de la glutathion réductase et de la superoxyde dismutase dans les cellules, rendant ainsi la juglone efficace à une dose plus faible. Nous montrons également que la combinaison juglone-ascorbate supprime la migration cellulaire, l'invasion et l'expression de gènes promoteurs de tumeurs et angiogéniques dans la lignée ARO, perturbant ainsi la capacité de transition épithélio-mésenchymateuse des cellules. Dans l’ensemble, nous montrons que l’acide ascorbique augmente le pouvoir cytotoxique de la juglone par le biais du cycle redox lorsqu’il est utilisé en synergie.
