La production de plantes haploïdes doublées (DH) est un outil efficace dans les programmes de sélection génétique et de sélection végétale; cependant, le poivron (Capsicum annuum L.) est récalcitrant à l'embryogenèse des microspores et à la production de DH. En essayant de briser la barrière de la production de DH, trois expériences indépendantes ont été menées sur l’embryogenèse de microspores de poivron doux. Dans la première expérience, l’effet des prétraitements au froid (4 ° C) et à la chaleur (32 ° C) a été étudié sur l’embryogenèse de microspores de trois génotypes de poivron comprenant les cultivars "Inspiration F1", "Maratus F1" et "Magno F1". dans une conception factorielle avec trois réplications. Le choc thermique (32 ° C pendant 7 jours), appliqué aux anthères «Inspiration F1» dépourvues de mannitol, a montré un pourcentage de microspores multinucléaires plus élevé, un nombre de structures multicellulaires, d'embryons totaux, d'embryons cotylédonaires et de régénérants. Dans la deuxième expérience, l’effet de différentes concentrations de putrescine (0, 0,5, 1, 2 et 5 mg l (-1)) a été évalué sur l’embryogenèse de microspores des trois cultivars de poivron susmentionnés. Le nombre moyen le plus élevé de structures multicellulaires, d'embryons cotylédonaires et de régénérants a été atteint en appliquant 0,5 à 1 mg de l (-1) de putrescine pendant les traitements de privation de mannitol et de choc thermique (32 degrés C) de cultures de microspores isolées du cultivar "Inspiration F1" . Une diminution significative de l'efficacité de l'embryogenèse des microspores a été observée lorsque des taux élevés de putrescine (2 et 5 mg l (-1)) ont été utilisés. L'embryogenèse des microspores a été complètement évitée à 5,0 mg de l (-1) de putrescine. Dans la troisième expérience, l’effet de différentes concentrations d’acide ascorbique (0, 20, 50, 100 et 200 mg 1 (-1)) a été étudié et les résultats ont montré que l’application d’acide ascorbique (20 et 50 mg 1 (-1)) -1)) au cours de la famine au mannitol et du traitement de choc thermique (32 ° C) a entraîné une amélioration remarquable du nombre d'embryons cotylédonaires produits et de leur capacité de régénération par rapport au traitement témoin. Cependant, l'application de taux plus élevés d'acide ascorbique (100 et 200 mg l (-1)) a inhibé les divisions cellulaires des microspores et l'embryogenèse. En conclusion, les résultats ont montré que la putrescine et l'acide ascorbique ont un effet significatif sur l'efficacité de l'embryogenèse des microspores du poivron doux lorsqu'ils sont utilisés à des concentrations appropriées.
