Effets anti-âge d'un nouveau sérum facial contenant de l'acide L-ascorbique, des protéoglycanes et un tripeptide stimulant les protéoglycanes: études sur explants de peau ex vivo et études cliniques in vivo sur des femmes.

Contexte: Avec l’âge, la diminution des taux de protéoglycanes, de collagène et d’élastine dans le derme entraîne l’apparition d’une peau vieillie. L'oxydation, largement motivée par des facteurs environnementaux, joue un rôle central. Objectif: Le but de cette étude était d'évaluer l'efficacité anti-vieillissement d'un sérum topique contenant de l'acide L-ascorbique, des protéoglycanes solubles, de l'acide hyaluronique de faible poids moléculaire et un tripeptide dans des études cliniques ex vivo et in vivo. Méthodes: Le photovieillissement et les dommages photo-oxydants ont été induits dans des explants de peau humaine par un rayonnement solaire artificiel. Les marqueurs de stress oxydatif - espèces réactives de l'oxygène (ROS), glutathion total (GSH) et dimères de cyclobutane pyrimidine (CPD) - ont été mesurés dans des explants traités au sérum et des témoins non traités. Le vieillissement chronologique a été simulé à l'aide d'hydrocortisone. Dans les deux études ex vivo, le collagène, l'élastine et les protéoglycanes ont été déterminés comme mesures de la dégradation de la matrice cutanée. Chez les femmes âgées de 21 à 67 ans, l'hydratation a été mesurée jusqu'à 24 heures après une seule application de sérum, à l'aide d'un cornéomètre et d'un hygromètre. Les perceptions des sujets concernant l'efficacité et l'acceptabilité ont été évaluées via un questionnaire après une application de sérum une fois par jour pendant 4 semaines. Les études ont été réalisées sous la supervision d'un dermatologue. Résultats: Dans l’étude de photovieillissement, l’irradiation a induit des modifications des taux de ROS, de CPD, de GSH, de collagène et d’élastine; ces changements ont été inversés par l'application de sérum topique. Le sérum protège également contre la réduction induite par l'hydrocortisone des taux de collagène, d'élastine et de protéoglycanes, qui étaient significativement plus élevés dans le groupe traité au sérum par rapport aux explants non traités à l'hydrocortisone. Dans les études cliniques, l’application de sérum a considérablement augmenté l’humidité de la peau pendant 6 heures. Des volontaires en bonne santé ont perçu le produit comme efficace pour rendre la peau plus lumineuse, plus hydratée et réduire les rides, et souhaitaient continuer à l'utiliser. Le sérum était bien toléré et non comédogène. Conclusion: le sérum protégé contre les dommages oxydatifs et la perte de protéines dermiques par le vieillissement photo et chronologique dans les explants de peau humaine. In vivo, le sérum a hydraté la peau pendant 6 heures et les utilisateurs ont perçu une augmentation de la luminosité et de l'hydratation de la peau, ainsi qu'une diminution des rides.
